Zheng Dong et al.

Summary: The complete genome sequence of peony leafroll-associated virus (PLRaV) was determined and its genetic characteristics were analyzed. The study found that PLRaV shares similarities with other members of the genus Ampelovirus in terms of genome structure and sequence. Phylogenetic analysis showed that PLRaV belongs to subgroup I of Ampelovirus.

Functional Characterization of Replication-Associated Proteins Encoded by Alphasatellites Identified in Yunnan Province, China

Liling Zhao et al.

Summary: Alphasatellites associated with begomovirus/betasatellite complexes have been found to modulate disease symptoms and viral DNA accumulation. This study revealed that alpha-Reps have the ability to suppress PTGS and reverse TGS, and they can also enhance the accumulation of helper virus. These findings suggest that alpha-Reps may play a role in overcoming host defense.

Geminiviruses employ host DNA glycosylases to subvert DNA methylation-mediated defense

Xiaojian Gui et al.

Summary: DNA methylation plays important roles in gene regulation and transposon silencing. This study reveals that active DNA demethylation can target viral DNAs and promote their virulence. The beta C1 protein of tomato yellow leaf curl China virus interacts with plant DNA glycosylases to promote demethylation of viral DNA and enhance viral virulence.

BmNPV p35 Reduces the Accumulation of Virus-Derived siRNAs and Hinders the Function of siRNAs to Facilitate Viral Infection

Shudi Zhao et al.

Summary: Antiviral immunity involves RNA interference (RNAi) pathway, and viruses can evade this defense by encoding viral suppressors of RNAi (VSRs). We found that p35 of BmNPV disrupts siRNA function and prevents their loading into Ago2, enhancing viral replication.

The plant protein NbP3IP directs degradation ofRice stripe virusp3 silencing suppressor protein to limit virus infection through interaction with the autophagy-related protein NbATG8

Liangliang Jiang et al.

Summary: A newly identified plant protein, NbP3IP, has been shown to degrade the silencing suppressor protein p3 of RSV through the autophagy pathway, thereby inhibiting RSV infection. The interaction of NbP3IP with autophagy proteins suggests a potential role in selective autophagosomal cargo receptor activity. This study provides evidence that autophagy can play an antiviral role against negative-strand RNA viruses by antagonizing p3 of RSV.

Recent advances in understanding plant antiviral RNAi and viral suppressors of RNAi

Yun Jin et al.

Summary: Molecular interactions between plants and viruses provide insights into host antiviral immunity and viral counter-defense mechanisms. Antiviral defense in plants is initiated by virus-derived small-interfering RNAs, while plant viruses have evolved viral suppressors of RNAi to counteract this defense. Recent research has shown that plant antiviral responses are regulated by endogenous small silencing RNAs, RNA decay, and autophagy, with some viral suppressors also targeting these defense responses to promote infection.

A calmodulin-binding transcription factor links calcium signaling to antiviral RNAi defense in plants

Yunjing Wang et al.

Summary: RNA interference (RNAi) is a gene regulatory and defense mechanism activated through calcium signaling, involving RNA-dependent RNA polymerase and Bifunctional nuclease to combat virus intrusion. Knockdown/knockout of Ca2+-related genes increases susceptibility to various viruses, indicating the importance of these genes in plant antiviral defense. Geminivirus V2 protein disrupts the calmodulin-CAMTA3 interaction to counteract RNAi defense, showing the versatility of host antiviral defense and viral counter-defense.

Viral suppressors from members of the family Closteroviridae combating antiviral RNA silencing: a tale of a sophisticated arms race in host-pathogen interactions

Muhammad Dilshad Hussain et al.

Summary: RNA silencing is an important mechanism for plant antiviral immunity, triggered by vsiRNAs derived from viruses. Viral suppressors of RNA silencing (VSRs) have evolved to combat plant antiviral responses, highlighting the arms race between plants and viruses. Family Closteroviridae encode VSRs to ensure successful infections in plants, showcasing the molecular strategies of virus-host interactions.
