Engineered yeast for efficient de novo synthesis of 7-dehydrocholesterol

Summary: In this study, a gene co-expression network was constructed for Saccharomyces cerevisiae with different nitrogen sources, leading to the identification of a key gene co-expression module related to nitrogen source preference utilization. Functional studies revealed the regulatory roles of endocytosis-related genes and mitochondria-related genes in the utilization of non-preferred amino acids and the reduction of urea accumulation. Furthermore, the study highlighted the potential positive effects of endocytosis and mitochondrial ribosomes protein translation on nitrogen source preference.

Enhancing Squalene Production in Saccharomyces cerevisiae by Metabolic Engineering and Random Mutagenesis

Summary: This study aimed to improve squalene production by metabolic engineering and random mutagenesis. The mevalonate pathway was enhanced and the beta-oxidation pathway was enhanced with multiple copies of key genes. A high throughput screening strategy was also established. These findings may facilitate the development of industrial squalene production by fermentation and potentially, other terpenoids.

Metabolic Engineering of Saccharomyces cerevisiae for Ethyl Acetate Biosynthesis

Introduction of heterologous AATase further boosted the synthesis of ethyl acetate. Sterol Composition Modulates the Response of Saccharomyces cerevisiae to Iron Deficiency

Summary: Iron deficiency affects iron uptake and growth in yeast cells, while changes in ergosterol biosynthesis patterns impact the response to iron deprivation. Mislocalization of the iron-sensing factor Aft1 to the vacuole disrupts its transcriptional activation function. These findings reveal novel connections between iron and sterol metabolisms with implications for yeast-derived food production and treatment of fungal infections with ergosterol biosynthesis-targeting drugs.
