Epidemiology of osteoarthritis

Summary: This narrative review summarizes the current evidence on the prevalence, incidence, and risk factors of osteoarthritis (OA) at the person-level and joint-level. The review highlights the high prevalence of OA worldwide, with older individuals, women, certain racial and ethnic groups, and individuals with lower socioeconomic status being more affected. Obesity and joint injury are identified as modifiable risk factors with the strongest evidence. Other emerging topics with potential associations with OA risk or progression include specific vitamins and diets, high blood pressure, genetic factors, metformin use, bone mineral density, abnormal joint shape and malalignment, and lower muscle strength/quality. The review also emphasizes the heterogeneity of OA and the importance of understanding and defining OA phenotypes. Overall, OA is a prevalent condition with significant global impacts on health outcomes, necessitating efforts to mitigate obesity and joint injury risks.

L-Gln up-regulates the expression of long non-coding RNA NKILA, inhibits the activity of relevant proteins, decreases the production of inflammatory mediators, and reduces the degradation of extracellular matrix, thereby alleviating the symptoms of arthritis.

IκB-ζ signaling promotes chondrocyte inflammatory phenotype, senescence, and erosive joint pathology

Summary: Osteoarthritis is a joint disease characterized by the degradation of cartilage and loss of joint mobility. This study found that chondrocytes from osteoarthritic mice and patients express pro-inflammatory markers and RANKL in a manner dependent on NF-kappa B. Exposure of chondrocytes to IL-1 beta and bone particles, which mimic bone matrix breakdown products in synovial fluid, recapitulated the inflammatory phenotype of chondrocytes. Bone particles and IL-1 beta induced senescent and apoptotic changes in chondrocytes through oxidative stress from various cellular sources. These findings enhance our understanding of the inflammatory mechanisms involved in osteoarthritis.

Understanding chondrocyte metabolism and altered metabolic pathways can provide insights into age-related OA pathophysiology and guide the development of new therapeutic targets.
