MicroRNA sequence codes for small extracellular vesicle release and cellular retention

Summary: Research has shown that miRNAs have sorting sequences that determine their secretion in sEVs, different cell types preferentially use specific sorting sequences to define their sEV miRNA profile. Insertion or deletion of these sequences affects miRNA secretion. RNA-binding proteins Alyref and Fus are involved in the mechanism, leading to enhanced miRNA delivery and inhibition of target genes in distant cells.

Extracellular microRNA 3' end modification across diverse body fluids

Summary: MicroRNAs (miRNAs) are small non-coding RNAs that play crucial roles in gene regulation. A new bioinformatic method, miNTA, was developed to identify 3' non-templated additions (NTAs) of miRNAs in extracellular biofluids. The study revealed that 3' NTA levels in miRNAs can differentiate between different types of biofluids and these modifications may stabilize base-pairing between miRNAs and their target genes.

Summary: Although immune checkpoint inhibitors (ICIs) have improved overall survival for cancer patients, the optimal duration of therapy remains undefined and further research is needed. Early trials have shown that a significant percentage of patients have durable responses to ICIs, prompting the question of whether patients may be receiving too much treatment and if durable remissions can still be achieved with shorter courses. Designing well-controlled trials to identify biomarkers for predicting patients who would benefit from shorter courses of immunotherapy is crucial for future work in this area.

MiRNA post-transcriptional modification dynamics in T cell activation

Summary: T cell activation results in changes in the miRNA repertoire, with some individual miRNAs being specifically upregulated. Using next-generation sequencing, differential expression of multiple miRNAs not previously linked to T cell activation was identified. Upregulated miRNAs showed a higher frequency of 3' adenylation, which may serve a protective function in the adverse environment and could be utilized to improve miRNA stability for T cell-targeted therapy.
