Inhibiting Mycobacterium abscessus Cell Wall Synthesis: Using a Novel Diazabicyclooctane beta-Lactamase Inhibitor To Augment beta-Lactam Action

Summary: Mycobacterium abscessus (Mab) infections pose a growing threat to patients, especially those with lung diseases and cystic fibrosis. Finding effective combinations of beta-lactam antibiotics and beta-lactamase inhibitors to disrupt cell wall synthesis in Mab is beneficial. The novel diazobicyclooctane beta-lactamase inhibitor durlobactam (DUR) enhances the susceptibilities of Mab isolates to beta-lactams in cell-based assays. DUR potently inhibits the beta-lactamase Bla(Mab) and forms stable acyl-enzyme complexes, providing a biochemical rationale for its activity.

Spatiotemporal localization of proteins in mycobacteria

Summary: This study systematically defined the subcellular organization of mycobacteria through high-throughput imaging and a customized computational pipeline, revealing a spatial organization network of over 700 conserved mycobacterial proteins as well as previously uncharacterized cell-cycle-dependent dynamics of essential proteins. The results provide a systems perspective on mycobacterial subcellular organization and tools for analyzing bacteria with non-standard growth characteristics.

Modeling Site-Specific Nucleotide Biases Affecting Himar1 Transposon Insertion Frequencies in TnSeq Data Sets

Summary: The study found that there are site-specific biases affecting the frequency of insertion of the Himar1 transposon at different TA sites. By developing a quantitative model based on patterns in the nucleotides surrounding TA sites, it is possible to predict expected counts at each site and improve estimates of gene fitness effects. The TTN-Fitness method allows for finer distinctions among essential and nonessential genes by comparing observed and expected counts based on insertion preferences.

Stepwise pathogenic evolution of Mycobacterium abscessus

Summary: The study suggests that epigenetic modifiers acquired through horizontal gene transfer can lead to sudden increases in the pathogenic potential of specific environmental clones, while allopatric parallel evolution during chronic lung infection promotes rapid increases in virulence. The findings indicate that constrained pathogenic evolution is observed while person-to-person transmission remains indirect.
