A protocol to assess cellular bioenergetics in flavivirus- infected cells

John Z. H. Low et al.

Summary: Aberrant cellular bioenergetics can have detrimental consequences in host cells, such as disrupting cellular bioenergetics and leading to apoptosis. This article describes methods for flavivirus propagation, titering, and infection, as well as procedures for stress tests on mitochondria and glycolysis.

TMEM41B Is a Pan-flavivirus Host Factor

H. Heinrich Hoffmann et al.

Summary: Flaviviruses pose a constant threat to human health, being transmitted by infected mosquitoes and ticks, and causing regular outbreaks. Through full-genome loss of function CRISPR-Cas9 screens, TMEM41B and VMP1 were identified as key factors in flavivirus infection. TMEM41B was found to be essential for all tested members of the Flaviviridae family, as well as for SARS-CoV-2 of the Coronaviridae. Single nucleotide polymorphisms in nearly 20% of East Asian populations were shown to reduce flavivirus infection.

Nuclear dengue virus NS5 antagonizes expression of PAF1-dependent immune response genes

Marine J. Petit et al.

Summary: The study reveals that PAF1 is involved in restricting DENV replication and promoting the expression of immune response genes, while the interaction between NS5 and PAF1C leads to the suppression of PAF1-dependent gene expression. These findings shed light on the role of PAF1C in DENV infection.

Genome-scale CRISPR screen identifies TMEM41B as a multi-function host factor required for coronavirus replication

Limeng Sun et al.

Summary: Our study identified potential therapeutic targets for common RNA viruses and revealed that targeting TMEM41B is a highly promising approach for the development of broad-spectrum anti-viral therapeutics. The KO of TMEM41B can strongly inhibit viral infection and delay the progression of a CoV disease, indicating its critical role in the replication cycle of coronaviruses.

Summary: Mitophagy and zymophagy are selective autophagy pathways induced in acute pancreatitis, with mitochondrial dysfunction and changes in mitochondrial dynamics observed. A novel DRP1-Parkin1-VMP1 selective autophagy pathway was identified during acute pancreatitis, mediating the selective degradation of damaged mitochondria and providing insights for potential therapeutic strategies.

TMEM41B and VMP1 are phospholipid scramblases

Tizhong Zhang et al.

Summary: TMEM41B and VMP1, two endoplasmic reticulum-resident transmembrane proteins, play crucial roles in regulating the formation of lipid droplets, autophagy initiation, and viral infection by affecting cholesterol and phosphatidylserine distribution. Their scramblase activity is key to understanding their mechanism of action.

TMEM41B is a host factor required for the replication of diverse coronaviruses including SARS-CoV-2

Joseph D. Trimarco et al.

Summary: The ER-resident host protein TMEM41B has been identified as an essential factor for multiple coronaviruses, including HCoV-229E and SARS-CoV-2. It likely contributes to viral replication complex formation by mobilizing cholesterol and other lipids to facilitate host membrane expansion and curvature. Targeting TMEM41B and its associated pathways may lead to the development of broad-spectrum anti-coronavirus therapeutics.
