Signaling pathways regulating the fate of fibro/adipogenic progenitors (FAPs) in skeletal muscle regeneration and disease

Giulio Giuliani et al.

Summary: Fibro/adipogenic progenitor cells (FAPs) play a critical role in modulating muscle regeneration, supporting MuSCs in maintaining tissue homeostasis, but disruption of niche cues can lead to their differentiation into harmful adipocytes and fibroblasts. Efforts are focused on understanding the molecular circuits governing their behavior and utilizing single-cell technologies to study FAP transitional states in muscle regeneration and myopathies.

Summary: Treg cell plasticity plays a significant role in autoimmunity and cancer, characterized by loss of Foxp3 expression. It has great therapeutic potential through destabilizing Treg cells to promote anti-tumor immunity or enhancing Treg cell stability to attenuate chronic inflammation.

Tissue regulatory T cells: regulatory chameleons

Andres R. Munoz-Rojas et al.

Summary: T-reg cells located in non-lymphoid tissues exhibit distinct transcriptomes, T cell receptor repertoires, and dependencies on growth factors which allow them to function effectively in their home tissue. These cells play crucial roles in tissue homeostasis, metabolism, tissue repair, and regulating the fate of non-lymphoid cell progenitors. Their specialized functions are influenced by transcriptional programs which differ based on the tissue environment, providing potential targets for precision immunotherapeutics.
