Molecular Mechanisms Leading from Periodontal Disease to Cancer

Bartosz Kamil Sobocki et al.

Summary: Periodontitis is a prevalent condition that affects half of the adult population and has been associated with an increased risk of cancer. Understanding the molecular mechanisms underlying tumor development in periodontitis patients is of fundamental importance. This article presents literature and evidence on the involvement of Porphyromonas gingivalis and Fusobacterium nucleatum, two main oral pathogens in periodontitis, in promoting tumorigenesis. The potential links between the RANKL-RANK signaling axis, circulating cytokines/leukocytes, and carcinogenesis are also discussed. Clinical trials relevant to the topic are examined and their outcomes summarized.

Enterococcus faecalis OG1RF induces apoptosis in MG63 cells via caspase-3/-8/-9 without activation of caspase-1/GSDMD

Yang Li et al.

Summary: The study showed that Enterococcus faecalis OG1RF infection induced apoptosis in MG63 cells, increasing the percentage of apoptotic cells and lactate dehydrogenase release, activating the caspase-3/-8/-9 pathway, but not activating the pyroptotic pathway regulated by caspase-1/gasdermin D.

Roles of calpain in the apoptosis of Eimeria tenella host cells at the middle and late developmental stages

Xiao-Zhen Cui et al.

Summary: This study found that E. tenella infection can induce host cell apoptosis, and calpain regulates this process by modulating the expression of Bid and Bax. Inhibition of calpain activity can reduce the apoptotic rate and infection rate, suggesting its potential use for managing E. tenella infection in chickens during the middle and late developmental stages.

Lactobacilli Attenuate the Effect of Aggregatibacter actinomycetemcomitans Infection in Gingival Epithelial Cells

Manuela R. Bueno et al.

Summary: Probiotics may help achieve a balanced microbiome in periodontitis by interfering with the colonization of Aggregatibacter actinomycetemcomitans in the oral cavity. This in vitro study evaluated the effect of two commercially available lactobacilli strains on gingival epithelial cells infected with A. actinomycetemcomitans. The results suggest that these probiotics may reduce the ability of A. actinomycetemcomitans to interact with gingival epithelial cells and modulate the cells' response.

Microbiological analysis of endodontically treated teeth with apical periodontitis before and after endodontic retreatment

Marlos Barbosa-Ribeiro et al.

Summary: The study found that the microbiota of teeth with endodontic treatment failure is polymicrobial, with Enterococcus faecalis and Porphyromonas gingivalis predominating in all phases of retreatment. Specific bacteria were associated with certain clinical and radiographic features.

The benefit of culture-independent methods to detect bacteria and fungi in re-infected root filled teeth: a pilot study

H. Al-Sakati et al.

Summary: The study aimed to identify dominant microorganisms in root filled teeth with apical periodontitis using Pan-PCR compared to culture-dependent approaches, focusing on fungal species profiling. The results showed that gutta-percha samples had fewer bacterial taxa with a dominance of Streptococcus, and fungus-specific cultures were negative. Pan-PCR assays were effective in detecting uncultivable or difficult-to-cultivate microorganisms, presenting an advantage over conventional microbiological diagnostics.

Inflammasomes in Alveolar Bone Loss

Yang Li et al.

Summary: Bone remodeling is tightly controlled by osteoclast-mediated bone resorption and osteoblast-mediated bone formation, and dysregulated inflammasome activation may lead to pathological bone loss in the alveolar bone. This is associated with various diseases and understanding the mechanism of inflammasome activation is crucial in developing therapeutic strategies for alveolar bone loss.

Summary: The study compared the antimicrobial substantivity effect of 3.8% SDF with other commonly used endodontic irrigants such as 2% CHX and 6.25% NaOCl. Results showed that 3.8% SDF exhibited comparable substantivity to 2% CHX and significantly higher than 6.25% NaOCl.

Neuronal cell life, death, and axonal degeneration as regulated by the BCL-2 family proteins

James M. Pemberton et al.

Summary: Axonal degeneration and neuronal cell death are regulated by BCL-2 family proteins, with potential influences from other cell death pathways. Deciphering the subsets of proteins required and separately regulating these outcomes is essential for future research and pharmacological interventions.
