Macrophage Control of Incipient Bone Formation in Diabetic Mice

Miya Kang et al.

Summary: This study found that macrophages from diabetic mice exhibited an inflamed phenotype at resting state. Analysis showed that bone regeneration was relatively impaired in diabetic mouse calvaria and in C57BL/6J mouse calvaria supplemented with diabetic mouse macrophages. Additionally, conditioned medium from diabetic mice had a negative impact on osteogenic differentiation of mouse mesenchymal stem cells compared to C57BL/6J mice.

Modulating macrophage activities to promote endogenous bone regeneration: Biological mechanisms and engineering approaches

Yiming Niu et al.

Summary: The coordinated interaction between osteogenesis and the osteoimmune microenvironment is crucial for successful bone healing. Macrophages, as highly plastic cells, play a central regulatory role in bone repair by mediating host immune response and exerting regenerative potency based on the signals they sense. Various engineering strategies are being developed to recruit, activate, and modulate the phenotype transition of macrophages to enhance the performance of engineered bone tissue.

The multifaceted roles of macrophages in bone regeneration: A story of polarization, activation and time

Claudia Schlundt et al.

Summary: Macrophages are a highly plastic and multifunctional cell population found in all tissues of the human body. In bone, different subpopulations of macrophages play essential roles in the cascade of regeneration and reconstitution, participating in both the inflammatory phase and later phases of healing. The M1/M2 paradigm may not fully capture the dynamic plasticity of macrophages, and understanding the specific mechanisms driving macrophage function in impaired healing scenarios could lead to new therapeutic targets.
