Quantitative characterization of recombinase-based digitizer circuits enables predictable amplification of biological signals

Katherine A. Kiwimagi et al.

Summary: Kiwimagi & Letendre et al. present a workflow to quantitatively define recombinase-based digitizer and predict responses to different input signals. With a mechanistic/phenotypic model that can predict circuit performance, they generate a synthetic cell-cell communication device that amplifies a synNotch output signal.

Imaging of multiple fluorescent proteins in canopies enables synthetic biology in plants

Stephen B. Rigoulot et al.

Summary: Reverse genetics approaches have revolutionized plant biology and agriculture, the prospects of bridging plant phenotypes with genes are provided by phenomics, and genetically encoded fluorescent proteins have transformed plant biology paradigms. Despite the availability of a wide range of fluorescent proteins, modern phenomics has largely overlooked fluorescence as a transgene expression tool. The new platform, FILP, allows for stand-off imaging of plant canopies expressing a variety of FP genes, enabling rapid synthetic promoter screening and analysis of fluorescence signals in plant canopies.

Wheat with greatly reduced accumulation of free asparagine in the grain, produced by CRISPR/Cas9 editing of asparagine synthetase gene TaASN2

Sarah Raffan et al.

Summary: This study successfully edited wheat lines using CRISPR/Cas9 technology to knock out the asparagine synthetase gene TaASN2, leading to a significant reduction in free asparagine concentration in the grain. These edits affected the concentrations of free asparagine, glutamine, glutamate, and aspartate in seeds across multiple generations.

Synthetic promoter designs enabled by a comprehensive analysis of plant core promoters

Tobias Jores et al.

Summary: A massively parallel reporter assay was used to measure the activity of nearly complete sets of promoters from Arabidopsis, maize and sorghum in two assay systems, uncovering the sequence features affecting promoter strength and facilitating promoter strength prediction and synthetic design. The experiments revealed species-specific differences in the contributions of GC content and transcription factors to promoter strength.
