Phagocytosis by an HIV antibody is associated with reduced viremia irrespective of enhanced complement lysis

David A. Spencer et al.

Summary: Antibodies are increasingly used to treat and prevent viral infections. In this study, the researchers investigated the role of antibody-mediated effector and complement activity in protecting against SHIV in rhesus macaques. They found that enhancing complement function did not improve the protective efficacy of the antibody, suggesting that effector functions of the unmodified antibody contribute to its effectiveness.

Prolonged viral suppression with anti-HIV-1 antibody therapy

Christian Gaebler et al.

Summary: Immunotherapy with anti-HIV-1 antibodies has the potential to suppress infection and increase the rate of clearance of infected cells. A clinical study showed that 76% of HIV-infected individuals who received a combination of antibodies maintained virologic suppression for at least 20 weeks without antiretroviral therapy. The administration of antibodies affected the HIV-1 reservoir, but further research is needed to define the precise effect of antibody immunotherapy.

An exploration of how broadly neutralizing antibodies might induce HIV remission: the 'vaccinal' effect

Timothy Tipoe et al.

Summary: This review explores the mechanisms of action of broadly neutralizing antibodies (bNAbs) and summarizes the evidence supporting their immunomodulatory properties. The authors propose the concept of a 'vaccinal effect' where bNAbs lead to sustained viral remission by blocking viral entry and through immune-modulatory mechanisms. The potent antiviral efficacy of bNAb therapy is emerging, but the ability of bNAbs to induce sustained viral suppression below therapeutic levels remains controversial.

Global post-translational modification profiling of HIV-1-infected cells reveals mechanisms of host cellular pathway remodeling

Jeffrey R. Johnson et al.

Summary: Viruses remodel host cellular pathways to replicate and evade immune defenses and they do so with limited genomic coding capacity. This study used mass spectrometry-based proteomics to analyze protein abundance and post-translational modifications in response to HIV-1 infection. The results revealed the importance of Aurora kinase activity in HIV-1 infection and identified potential substrates of a degraded phosphatase during infection. Additionally, the study showed that the HIV-1 Vpr protein inhibits histone H1 ubiquitination, leading to DNA repair defects.

HIV-1 Reservoir Persistence and Decay: Implications for Cure Strategies

Edward F. Kreider et al.

Summary: Although antiretroviral therapy (ART) suppresses HIV-1 replication, a viral reservoir persists in individuals living with HIV. Recent studies have shown that the HIV-1 reservoir is dynamic, with expansion and contraction of virus-infected cells over time. Evidence suggests that host immunity exerts selective pressure on reservoir viruses and clears HIV-1 infected cells over years on ART. Therefore, strategies involving immunotherapy or genomic engineering are being developed to enhance reservoir suppression or directly eliminate intact viruses.

A guide to: generation and design of nanobodies

Serge Muyldermans

Summary: Nanobodies, a trademark of Ablynx, are single antigen-binding domains of heavy chain-only antibodies in Camelidae produced recombinantly for research and medical applications. Various methods are used for their generation, producing cost-effective Nbs with high affinity for their target antigens. Additionally, engineering technologies have been explored to broaden the application range of Nbs for potential advantages over other affinity reagents.

Accelerated Clearance and Degradation of Cell-Free HIV by Neutralizing Antibodies Occurs via FcγRIIb on Liver Sinusoidal Endothelial Cells by Endocytosis

James M. Turman et al.

Summary: The study demonstrates that neutralizing antibodies can suppress HIV infection by accelerating viral clearance from blood circulation. The Fc effector mechanism of liver sinusoidal endothelial cells (LSEC) Fc gamma RIIb-mediated endocytosis plays a crucial role in eliminating cell-free HIV, which has implications for the development of HIV vaccines and antibody therapies.

Inhalable Nanobody (PiN-21) prevents and treats SARS-CoV-2 infections in Syrian hamsters at ultra-low doses

Sham Nambulli et al.

Summary: The study demonstrates the high efficacy of PiN-21 in preventing and treating SARS-CoV-2 infection in Syrian hamsters modeling moderate to severe COVID-19. Intranasal delivery of PiN-21 effectively protects infected animals from weight loss and reduces viral burdens in both lower and upper airways.
