Computational Insights in DNA Methylation: Catalytic and Mechanistic Elucidations for Forming 3-Methyl Cytosine

Summary: Methylation at the C5 position of cytosine (5 mC) is a common epigenetic modification process linked to many human diseases. A recent discovery shows that DNA methyltransferases (DNMTs) can add a methyl group at position 3 to yield 3 mC, which can cause severe damages to DNA. By using density functional theory (DFT) modeling studies, we have gained insight into the mechanism of this new methylation approach and discovered an important off-target activity of DNMTs.
