Enantioselective hydroamination of unactivated terminal alkenes

In this study, an enantioselective and intermolecular hydroamination reaction of unactivated terminal alkenes was developed, which occurs with equimolar amounts of alkene and amine, shows high tolerance towards functional groups, and yields products with high enantioselectivity and turnover numbers.

This method involves regio-and stereoselective hydrometalation of unactivated alkenes with nontrivial Markovnikov selectivity, followed by cross-coupling with unactivated alkyl electrophiles to generate trialkyl tertiary saturated stereogenic centers. The mild and robust conditions allow for the use of different types of unactivated alkenes and alkyl electrophiles, resulting in diverse trialkyl tertiary stereogenic carbon centers with broad functional group tolerance.

By tuning olefin isomerization and migratory hydroamination, gamma-selective amination can be achieved via stabilization of a 6-membered nickellacycle and subsequent interception by an aminating reagent. This approach enables late-stage access to value-added products and has been further extended to delta-selective amination by employing picolinamide-coupled alkene substrates.

An umpolung-enabled copper-catalysed regioselective hydroamination approach to α-amino acids

Summary: This study established a copper-catalysed hydroamination method for the regio- and stereoselective synthesis of alpha-amino acids from acrylates using hydrosilanes and hydroxylamines. Regioselectivity control was achieved by utilizing hydroxylamine as an electrophilic amination reagent. The use of CsOPiv base and DTBM-dppbz ligand facilitated the challenging C-N bond formation at the alpha position, while the chiral ligands Xyl-BINAP and DTBM-SEGPHOS controlled the stereochemistry at the beta-position. Additionally, the chiral auxiliary (-)-8-phenylmenthol induced stereoselectivity at the alpha-position to produce optically active unnatural alpha-amino acids with adjacent stereocentres.
