Breaking constraint of mammalian axial formulae

Gabriel M. Hauswirth et al.

Summary: Vertebral column length and shape are highly conserved within a species, but show diversity across species. This study reveals the molecular mechanisms that constrain vertebral number in mice and highlights a novel role for posterior Hox genes in this process.

BAF complex-mediated chromatin relaxation is required for establishment of X chromosome inactivation

Andrew Keniry et al.

Summary: In this study, a replenishable female mouse embryonic stem cell system was developed to study X chromosome inactivation (XCI). Through a targeted genetic screen, the researchers identified the BAF complex as essential for creating nucleosome-depleted regions at promoters on the inactive X chromosome during the early stages of XCI establishment. This model system provides a valuable tool for discovering unknown factors involved in XCI and epigenetic silencing.

Maternal SMCHD1 controls both imprinted Xist expression and imprinted X chromosome inactivation

Iromi Wanigasuriya et al.

Summary: Embryonic development relies on proteins provided by the maternal oocyte, including factors that regulate DNA methylation. Maternal SMCHD1 is found to regulate both autosomal imprinted gene expression and X chromosome inactivation in mouse preimplantation embryos. This study expands the understanding of SMCHD1's role in enforcing the silencing of Polycomb targets.

HOX epimutations driven by maternal SMCHD1/LRIF1 haploinsufficiency trigger homeotic transformations in genetically wildtype offspring

Shifeng Xue et al.

Summary: This study reveals that Smchd1 regulates Hox gene expression at an epigenetic level in mammals, and its loss causes precocious and ectopic Hox transcription. The loss of maternal Smchd1 also leads to Hox epimutations and affects the vertebral patterning in offspring. Furthermore, the regulation is mediated by the direct interacting partner Lrif1.

BAP1 constrains pervasive H2AK119ub1 to control the transcriptional potential of the genome

Nadezda A. Fursova et al.

Summary: Histone modifications play crucial roles in gene regulation and development, with little known about modifications found broadly throughout the genome. BAP1 deubiquitylase constrains H2AK119ub1 accumulation throughout the genome, impacting gene expression and transcription. Failure to constrain H2AK119ub1 compromises Polycomb complex occupancy at certain target genes, resulting in their derepression.

H2AK119ub1 guides maternal inheritance and zygotic deposition of H3K27me3 in mouse embryos

Hailiang Mei et al.

Summary: The study reveals the preceding dynamics of H2AK119ub1 over H3K27me3 at the maternal-to-zygotic transition, and identifies PCGF1/6-vPRC1 as an essential player in maternal epigenetic inheritance.

Zhiyuan Chen et al.

Summary: In mouse preimplantation embryos, H2AK119ub1 and H3K27me3 have distinct genome-wide dynamics. Loss of H2AK119ub1 results in premature activation of developmental genes, while loss of H3K27me3 does not.
