Comparative metabolomics with Metaboseek reveals functions of a conserved fat metabolism pathway in C. elegans

Summary: In this study, untargeted metabolomics using high-resolution mass spectrometry and Metaboseek software was applied to uncover the biosynthetic functions of a fat metabolism pathway in C. elegans. Over 200 previously uncharacterized metabolites related to the alpha-oxidation pathway were identified, highlighting the importance of alpha-oxidation in fatty acid and lipid metabolism.

clinker & clustermap.js: automatic generation of gene cluster comparison figures

Summary: Genes involved in biological pathways are often found in gene clusters, and comparing these clusters can provide valuable insights into their function and evolutionary history. To simplify the tedious process of comparing and visualizing gene cluster similarity, a Python tool called clinker and a companion JavaScript library called clustermap.js have been developed to automatically generate accurate, interactive, publication-quality gene cluster comparison figures from sequence files.

Summary: Fungi maintain the balance of metal micronutrient pools through various homeostasis mechanisms. Metabolites derived from the xan biosynthetic gene cluster in Aspergillus fumigatus bind copper, affect cellular copper content, and exhibit significant metal-dependent antimicrobial properties. These metabolites also demonstrate copper-dependent antimicrobial properties against a variety of human pathogenic fungi and bacteria.

Secreted Secondary Metabolites Reduce Bacterial Wilt Severity of Tomato in Bacterial-Fungal Co-Infections

Summary: The study found that co-infection with Ralstonia solanacearum and Fusarium oxysporum can reduce the severity of bacterial wilt disease in tomato plants, with bikaverin synthesis by Fusarium contributing to this effect. The arrival time of each microbe at the infection court also impacts the severity of wilt disease. Additionally, analysis of the root secretome in co-infection revealed previously uncharacterized secreted metabolites that inhibit the growth of R. solanacearum and provide protection against bacterial wilt disease in tomato seedlings.

Summary: Fungi produce secondary metabolites that help them adapt to extreme environments, such as conidial pigment or melanin protecting them from extreme radiation. This study investigated the protective role of pigment produced by P. variotii against radiation, utilizing CRISPR-Cas9 mediated gene deletion to confirm its mechanism of action. Further identification of the pigment synthesized by the PKS gene of P. variotii contributes to understanding radiation resistance mechanisms.

Combinatorial Assembly of Modular Glucosides via Carboxylesterases Regulates C. elegans Starvation Survival

Summary: MOGLs, derived from combinatorial assembly of moieties from amino acid, neurotransmitter, and lipid metabolism, form a large metabolite library in the model organism C. elegans. The carboxylesterase homologue Cel-CEST-1.2 is responsible for specific 2-O-acylation of diverse glucose scaffolds, resulting in over 150 different MOGLs. Deletion of Cel-cest-1.2 leads to early death of adult animals under starvation conditions, revealing the biological functions of MOGLs.
