Edaravone activates the GDNF/RET neurotrophic signaling pathway and protects mRNA-induced motor neurons from iPS cells

Qian Li et al.

Summary: This study presents a method to facilitate iPSC differentiation and disease modeling. It shows that the FDA-approved drug edaravone has a neuroprotective effect and can enhance its clinical efficacy by activating the GDNF/RET neurotrophic signaling pathway. Furthermore, the combination treatment of edaravone and GDNF demonstrates better protection against neurotoxicity, suggesting a potential combination strategy for ALS treatment.

Failure of Glial Cell-Line Derived Neurotrophic Factor (GDNF) in Clinical Trials Orchestrated By Reduced NR4A2 (NURR1) Transcription Factor in Parkinson's Disease. A Systematic Review

Piniel Alphayo Kambey et al.

Summary: Parkinson's disease is a common neurodegenerative disorder with unknown etiology, making it challenging to develop effective therapies. Glial cell line-derived neurotrophic factor (GDNF) has shown promise in preclinical studies but has yielded equivocal clinical results. Recent research suggests that the failure of GDNF in clinical trials may be due to a deficiency in nuclear receptor-related factor 1 (Nurr1), which presents a potential therapeutic target for PD.

Expanding the Toolkit for In Vivo Imaging of Axonal Transport

Andrew P. Tosolini et al.

Summary: This paper details protocols for assessing in vivo axonal transport in mice, including distinguishing between motor and sensory neurons to analyze different neuronal subtypes, and evaluating the transport of signaling endosomes and mitochondria simultaneously. These complementary intravital approaches facilitate quantitative monitoring of axonal transport in health and disease by imaging different cargoes in distinct peripheral nerve axons.
