Microbial Metabolites Orchestrate a Distinct Multi-Tiered Regulatory Network in the Intestinal Epithelium That Directs P-Glycoprotein Expression

Dynamics of the normal gut microbiota: A longitudinal one-year population study in Sweden

Summary: The composition and function of the gut microbiota vary significantly between individuals, and this variation can potentially be explained by intra-individual dynamics. The abundance of Faecalibacterium prausnitzii and Bifidobacterium species is negatively associated with the stability of the gut microbiota composition. In contrast, facultative anaerobes and aerotolerant bacteria, such as Escherichia coli and Lactobacillus acidophilus, show extensive variation independent of compositional stability. The contribution of intra-individual variance to the total variance is greater for functional pathways than for microbial species. Therefore, reliable quantification of microbial features requires repeated sampling to address intra-individual variations of the gut microbiota.

Summary: The study finds that specific genera within the Clostridia and Bacilli classes of the intestinal microbiome are essential for inducing P-glycoprotein (P-gp) expression in the intestinal epithelium. Metabolites produced by these microbial communities, such as short-chain fatty acids and secondary bile acids, positively associate with P-gp expression and synergistically upregulate P-gp function. In patients with ulcerative colitis, reduced P-gp expression is linked to a decrease in anti-inflammatory endocannabinoids and luminal content that can induce P-gp expression, highlighting the important role of the microbiome in regulating intestinal inflammation.
