Summary: The study found that silencing Zona incerta (ZI) significantly blocked the expression of social conditioned place aversion induced by repeated social defeat stress (RSDS) while enhancing anxiety levels in mice experiencing RSDS. Additionally, silencing ZI altered PTSD-associated behaviors, such as promoting spatial cognitive impairment and preventing deficits in sensorimotor gating and social avoidance behavior. These effects were not observed in non-stressed mice. In summary, the results suggest ZI plays an important role in modulating RSDS-induced social CPA and PTSD-like behaviors.

Activation of somatostatin (SOM)-expressing neurons induces anxiety, while activation of calretinin (CR)-positive cells and inhibition of vesicular glutamate transporter-2 (Vglut2)-expressing neurons produce anxiolysis. These findings provide experimental evidence for the role of ZI subpopulations in processing anxiety.
