Low-intensity pulsed ultrasound promotes angiogenesis via the AKT pathway and DNA methylation in human umbilical vein endothelial cells

JiaYi Li et al.

Summary: The study demonstrated that LIPUS could promote the proliferation and angiogenic differentiation of endothelial cells (ECs) via the activation of the AKT signaling pathway. Additionally, LIPUS could alter the DNA methylation of ECs through the AKT signaling pathway activation, which in turn affected biomolecular changes in the cells.

Longitudinal effects of low-intensity pulsed ultrasound on osteoporosis and osteoporotic bone defect in ovariectomized rats

Shuxin Sun et al.

Summary: The study used two intensity levels of LIPUS, 30 mW/cm² and 150 mW/cm², to treat osteoporosis and osteoporotic bone defect in OVX rats. Results showed that 150 mW/cm² LIPUS led to faster healing of the bone defect compared to 30 mW/cm² after 3 weeks, but did not further enhance healing after 6 weeks. Additionally, 150 mW/cm² LIPUS had more advantages in improving bone density, microstructure, and biomechanics for osteoporosis compared to 30 mW/cm² after 6 weeks.

A glaucoma- and ALS-associated mutant of OPTN induces neuronal cell death dependent on Tbk1 activity, autophagy and ER stress

Swetha Medchalmi et al.

Summary: Mutations in OPTN, such as the 2-bp insertion, can lead to aberrant protein sequences, causing diseases like glaucoma and ALS. This mutation induces cell death, localizes mainly in the nucleus, and activates ER stress through Tbk1.

ASIC1a is required for neuronal activation via low-intensity ultrasound stimulation in mouse brain

Jormay Lim et al.

Summary: Accumulating evidence suggests that transcranial low-intensity ultrasound could be a non-invasive neural modulation tool for treating brain diseases. Low-intensity ultrasound can activate neurons in the mouse brain and induce adult neurogenesis in specific regions. ASIC1a is identified as the molecular determinant involved in the mechano-signaling of low-intensity ultrasound for neural activation in the mouse brain.

Development and validation of low-intensity pulsed ultrasound systems for highly controlled in vitro cell stimulation

F. Fontana et al.

Summary: This study describes the development and validation of two low-intensity pulsed ultrasound stimulation systems, which successfully control the dose delivered to biological targets. Through transducer characterization and carefully designed retaining systems, the pressure field shape and intensity were precisely controlled. Biological in vitro tests confirmed the sterility and safety of the system after multiple immersions in water.

Region-specific effects of ultrasound on individual neurons in the awake mammalian brain

Hua-an Tseng et al.

Summary: Using a high-resolution calcium imaging technique, researchers found that brief 100-ms-long ultrasound pulses can increase intracellular calcium in a large fraction of individual neurons in the hippocampus and the motor cortex of awake mice. The evoked calcium response in hippocampal neurons has a rapid onset with a latency shorter than 50 ms, and is shorter in duration and smaller in magnitude compared to that in the motor cortex. These results demonstrate that noninvasive ultrasound stimulation transiently increases intracellular calcium in individual neurons in awake mice, with brain region-specific response profiles.
