Expanding role of PI5P4Ks in cancer: A promising druggable target

Summary: Cancer cells adapt to microenvironmental stresses but also have vulnerabilities that can be targeted for therapy. PI5P4Ks, a family of stress-regulated kinases, are essential for metabolic adaptation in cancer cells and have a synthetic lethal interaction with the tumor suppressor p53. Understanding the role of PI5P4Ks in cancer signaling has led to the development of specific inhibitors for potential therapeutic interventions.

Phosphorylation of guanosine monophosphate reductase triggers a GTP-dependent switch from pro- to anti-oncogenic function of EPHA4

Summary: The signal transduction pathways governing post-translational regulation of nucleotide metabolism are poorly understood. This study reveals that phosphorylation of guanosine monophosphate reductase (GMPR) at Tyr267 is crucial for its activity and that phosphorylation by ephrin receptor tyrosine kinase EPHA4 decreases GTP pools. Furthermore, the study demonstrates that GMPR plays a key role in EPHA4-mediated suppression of RAC1, resulting in negative effects on melanoma cell invasion and tumorigenicity.

Development, synthesis, and biological evaluation of sulfonyl-α-l-amino acids as potential anti-Helicobacter pylori and IMPDH inhibitors

Summary: This study focused on the development and synthesis of sulfonyl-alpha-l-amino acids coupled with the anisamide scaffold for their anti-Helicobacter pylori and IMPDH inhibitory activities. Among the synthesized derivatives, compounds 6 and 10 exhibited promising inhibitory activities against the HpIMPDH enzyme, with IC50 values of 2.42 and 2.56 µM, respectively. Additionally, the compounds showed less activity and higher safety against hIMPDH2 compared to the reference drugs, indicating a potential selectivity towards HpIMPDH.

Inhibition of guanosine monophosphate synthetase (GMPS) blocks glutamine metabolism and prostate cancer growth

Summary: Glutamine is a critical nutrient in cancer, especially in prostate cancer; and Guanosine monophosphate synthetase (GMPS) plays a significant role in de novo purine biosynthesis pathway. High expression of GMPS is associated with decreased survival rates in prostate cancer, and inhibition or knockdown of GMPS can significantly decrease cell growth in prostate cancer cells. These findings highlight the importance of glutamine metabolism and suggest GMPS as a potential therapeutic target in prostate cancer.
