Common, low-frequency, rare, and ultra-rare coding variants contribute to COVID-19 severity

Chiara Fallerini et al.

Summary: The study developed a machine-learning model using common and rare exonic variants to predict COVID-19 severity. By selecting key Boolean features and combining them into an Integrated PolyGenic Score, the model offers insights into the genetic contribution to disease severity. A quarter of the selected genes are sex-specific, and pathway analysis highlighted the multi-organ nature of COVID-19 severity. This model could potentially aid in diagnostics, therapeutics, and disease management at the bedside.

Sialic acid-containing glycolipids mediate binding and viral entry of SARS-CoV-2

Linh Nguyen et al.

Summary: Evidence suggests that host glycans, specifically glycolipids containing sialic acid, play a role in facilitating the entry of SARS-CoV-2 virus into host cells by binding to the receptor-binding domain (RBD) of the spike protein. Depletion of cell surface sialic acid levels through various methods decreases RBD binding and infection of the virus, indicating the importance of sialylated glycans in viral entry.

EMDA: A Python package for Electron Microscopy Data Analysis

Rangana Warshamanage et al.

Summary: This article presents an open-source Python library called EMDA, which focuses on the validation of cryo-EM map and model manipulation. The library's functionalities are demonstrated through several examples, including the use of local correlation as a metric for identifying map-model differences and unmodeled regions. The mapping of local correlation to individual atoms and its insights on local signal variations are discussed. In addition, the likelihood-based map overlay and map magnification refinement in EMDA are demonstrated to emphasize their importance in structural comparison studies.

Binding of the SARS-CoV-2 spike protein to glycans

Wei Hao et al.

Summary: The study found that the spike proteins and subunits of SARS-CoV-2, SARS-CoV, and MERS-CoV can bind to heparan sulfate (HS) in a sulfation-dependent manner, with no binding to sialic acid residues detected. This suggests that HS binding may be a general mechanism for these coronaviruses to attach to host cells.

A potent SARS-CoV-2 neutralising nanobody shows therapeutic efficacy in the Syrian golden hamster model of COVID-19

Jiandong Huo et al.

Summary: Nanobodies engineered as homotrimers show promising potential in neutralizing SARS-CoV-2 viral strains for COVID-19 treatment, especially when administered through respiratory routes. The study characterizes four nanobodies with high affinity for the receptor binding domain of the spike protein, demonstrating efficacy in inhibiting disease progression in animal models.
