Second-Line Tisagenlecleucel or Standard Care in Aggressive B-Cell Lymphoma

M. R. Bishop et al.

Summary: Tisagenlecleucel did not demonstrate superiority over standard salvage therapy in this trial. Further studies are needed to evaluate which patients may benefit the most from each approach.

Axicabtagene Ciloleucel as Second-Line Therapy for Large B-Cell Lymphoma

F. L. Locke et al.

Summary: This international phase 3 trial showed that axicabtagene ciloleucel therapy significantly improved event-free survival and response in patients with early relapsed or refractory large B-cell lymphoma compared to standard care, despite the expected high-grade toxic effects.

Does bridging radiation therapy affect the pattern of failure after CAR T-cell therapy in non-Hodgkin lymphoma

Omran Saifi et al.

Summary: This study analyzed the pattern of disease failure after CART therapy for non-Hodgkin lymphoma and evaluated the local control rate and characteristics of in-field recurrences in patients who received bridging radiotherapy. The findings suggest that the majority of disease progressions after CART infusion involve pre-existing sites, and bridging radiotherapy prior to CART can provide excellent in-field local control and durable response.

Liver metastasis restrains immunotherapy efficacy via macrophage-mediated T cell elimination

Jiali Yu et al.

Summary: Liver metastases diminish the efficacy of immunotherapy by siphoning activated CD8(+) T cells from systemic circulation, creating systemic immune desert. Combining liver-directed radiotherapy with immunotherapy can promote systemic antitumor immunity.

Efficacy of Palliative Radiation Therapy (RT) for Chemotherapy Relapsed or Refractory Diffuse Large B-Cell Lymphoma: A Population-Based Retrospective Review

Jordan Wong et al.

Summary: Palliative radiation therapy is effective for symptom improvement in patients with diffuse large B-cell lymphoma. Factors such as R/R disease, not requiring concurrent steroids, response to first-line chemotherapy, and smaller lesion size can predict better treatment outcomes. There is no evidence to suggest that higher RT doses are more effective for palliation.

LYMPHOMA: ADVANCES IN IMAGING AND RADIOTHERAPY SPECIAL FEATURE: REVIEW ARTICLE The Current and Future Role of Radiation Therapy in the Era of CAR T- cell Salvage

Carl Deselm

Summary: Radiation therapy can modulate the immune system, playing a critical role in improving cancer patient outcomes when combined strategically with approved immunotherapies. The success of current cancer immunotherapies, particularly CAR T-cell therapy, highlights the potential for further advancements in the field.

Patterns and Predictors of Failure in Recurrent or Refractory Large B-Cell Lymphomas After Chimeric Antigen Receptor T-Cell Therapy

Nicholas B. Figura et al.

Summary: Most patients who recur after CAR T therapy experience a component of local progression. Lesions with high-risk features, particularly large size, are associated with inferior treatment efficacy and patient survival. Locally directed therapies to high-risk lesions, such as radiation therapy, may be a viable strategy to prevent CAR T failures in select patients.
