Sex Differences in Opioid and Psychostimulant Craving and Relapse: A Critical Review

Summary: This review suggests that there may not necessarily be a gender difference in vulnerability to psychostimulant and opioid craving and relapse, although imaging studies indicate potential sex differences in brain responses to drug cues and stress. Preclinical studies show evidence for sex differences in cocaine seeking reinstatement and incubation, but not in methamphetamine or opioid seeking reinstatement or incubation.

Sex differences in the effect of chronic delivery of the buprenorphine analogue BU08028 on heroin relapse and choice in a rat model of opioid maintenance

Summary: The study showed that chronic administration of BU08028 reduced the incubation of heroin seeking behavior, but unexpectedly may increase the re-acquisition of heroin self-administration in female rats. In male rats, BU08028 exhibited a selective decrease in extinction responding. Additionally, BU08028 had minimal effects on heroin choice behavior in both sexes.

Summary: The study found that in long access sessions, rats escalated their intake of fentanyl, with no reliable sex differences observed; however, in reinstatement tests, LgA rats were more likely to reinstate than ShA rats. Overall, there is a limited relationship between escalation of drug intake and craving in opioid use disorder.
