Rational Construction of an Artificial Binuclear Copper Monooxygenase in a Metal-Organic Framework

Summary: A novel strategy for constructing an artificial binuclear copper monooxygenase starting from a Ti metal-organic framework was reported. Experimental and computational studies showed that the enzyme exhibited outstanding catalytic activity in monoxygenation processes and significantly higher activity compared to a mononuclear copper monooxygenase. This was attributed to the cooperative stabilization of Cu-O-2 adduct by Cu-2 sites, leading to a lower free energy increase for O-O bond cleavage.

The basicity of an active-site water molecule discriminates between tyrosinase and catechol oxidase activity

Summary: The study showed that the presence of metallochaperone caddie protein (Cad) enables tyrosinase (Ty) to catalyze the catecholase reaction more effectively than the phenolase reaction. Mutations at the N191 residue can decrease the inherent ratio of phenolase versus catecholase activity. The increased basicity of the water molecule may be necessary for inhibiting the proton transfer in the catalytic cycle.
