Summary: Exogenous IBA is not necessary for ex vitro rooting in micropropagated shoots of Cedrela fissilis, and root induction is significantly reduced by auxin signaling and polar auxin transport inhibitors. The presence of meristematic cells and the formation of meristematic centers in the base of shoot cuttings at 3 days of rooting are necessary for adventitious root formation. Accumulation of cell division- and auxin-related proteins, as well as unique proteins related to cell wall modification, at 3 days of rooting are associated with higher IAA contents and AR initiation.

The combination of IBA with PG significantly increased root number and affected the abundance of proteins related to metabolic processes during rooting. This study demonstrates the potential of IBA and PG in promoting root development in difficult-to-root species, highlighting the importance of auxin signaling and growth-promoting properties of phenolic compounds in this process.
