Pharmacological inactivation of the prion protein by targeting a folding intermediate

Giovanni Spagnolli et al.

Summary: The researchers introduced a new drug discovery approach called Pharmacological Protein Inactivation by Folding Intermediate Targeting (PPI-FIT), which targets folding intermediates of disease-causing proteins. Testing this approach on the cellular prion protein, they identified ligands that could stabilize a folding intermediate and promote its degradation, supporting the idea that modulating protein levels by acting on their folding pathways may have therapeutic potential. Moreover, recent computational advancements in simulating biochemical processes have allowed for investigating protein regulation mechanisms with realistic physics-based models at an atomistic level.

Boceprevir, Calpain Inhibitors II and XII, and GC-376 Have Broad-Spectrum Antiviral Activity against Coronaviruses

Yanmei Hu et al.

Summary: The study identified four SARS-CoV-2 main protease inhibitors with potent antiviral activity and dual mechanism of action, targeting viral M-pro and host cathepsin L. These compounds showed broad-spectrum antiviral activity against SARS-CoV-2, SARS-CoV, MERS-CoV, and human coronaviruses 229E, OC43, and NL63. Additionally, the research demonstrated additive antiviral effect when these compounds were combined with remdesivir, suggesting their potential as promising starting points for further development against existing and emerging coronaviruses.

Comprehensive investigations revealed consistent pathophysiological alterations after vaccination with COVID-19 vaccines

Jiping Liu et al.

Summary: Large-scale COVID-19 vaccinations are currently underway, causing consistent alterations in hemoglobin A1c, serum sodium and potassium levels, coagulation profiles, and renal functions in healthy volunteers. Single-cell mRNA sequencing also revealed changes in gene expression of immune cells, suggesting that vaccination mimics infection. Increased NF-kappa B signaling and reduced type I interferon responses were observed, indicating caution is needed when vaccinating individuals with pre-existing clinical conditions.

Epigallocatechin gallate from green tea effectively blocks infection of SARS-CoV-2 and new variants by inhibiting spike binding to ACE2 receptor

Jinbiao Liu et al.

Summary: The study demonstrated that green tea beverage or its major ingredient, epigallocatechin gallate (EGCG), were highly effective in inhibiting live SARS-CoV-2 and human coronavirus infection, especially against new variants. Among the 4 active green tea catechins, EGCG showed the strongest antiviral effect by interfering with the binding of viral spikes to host cell receptors.
