The impact of individual human cytochrome P450 enzymes on oxidative metabolism of anticancer drug lenvatinib

Katarina Vavrova et al.

Summary: Lenvatinib, a small molecule tyrosine kinase inhibitor, shows good inhibitory effects in various carcinomas, especially in thyroid cancer. Metabolites of lenvatinib have been identified through in vitro studies using human hepatic microsomes and recombinant CYPs, with CYP3A4 and CYP1A1 playing significant roles in the metabolism of lenvatinib. Further research on drug-drug interactions and CYP modulators is needed for personalized therapy with TKIs.

Unraveling the Structural Basis of Selective Inhibition of Human Cytochrome P450 3A5

Jingheng Wang et al.

Summary: The human cytochrome P450 CYP3A4 and CYP3A5 enzymes play a significant role in metabolizing a large portion of marketed drugs. Differences in genetic polymorphism and tissue distribution between CYP3A4 and CYP3A5 can impact drug metabolism and toxicity during treatment. The discovery of selective inhibitors for CYP3A5, such as clobetasol propionate, provides a potential starting point for developing more potent and selective therapeutic options.
