Translational control through ribosome heterogeneity and functional specialization

David M. Gay et al.

Summary: The concept of ribosome specialization originates from the early days of molecular biology, but has recently gained momentum. Studies have identified distinct ribosome populations in various species and model systems, with some potentially having functional specialization in regulating specific mRNA translation. Further research in this field could reveal a new layer of gene expression control at the translation level, where the ribosome plays a key regulatory role.

The chaperone Tsr2 regulates Rps26 release and reincorporation from mature ribosomes to enable a reversible, ribosome-mediated response to stress

Yoon-Mo Yang et al.

Summary: This study reveals that under high Na+, sorbitol, or pH stress, a physiologically relevant ribosome population arises through the dissociation of Rps26 from fully assembled ribosomes, enabling a translational response to these stresses. The chaperone Tsr2 is involved in the release and reincorporation of Rps26, repairing the subunit after the stress subsides.

Regulation of translation by site-specific ribosomal RNA methylation

Martin D. Jansson et al.

Summary: Variation in ribosome composition due to differential 2'-O-methylation of ribosomal RNA can lead to ribosome heterogeneity, impacting translation of select mRNAs and resulting in changes in cellular phenotypes. This suggests a broader mechanism where specific regulation of rRNA modification patterns fine tunes translation.

Ribo-ODDR: oligo design pipeline for experiment-specific rRNA depletion in Ribo-seq

Ferhat Alkan et al.

Summary: The study revealed that rRNA fragments generated via Ribo-seq vary significantly with different experimental conditions, suggesting that a 'one-size-fits-all' approach may be inefficient. Therefore, they developed Ribo-ODDR, an oligo design pipeline integrated with a user-friendly interface to assist in selecting oligos for efficient experiment-specific rRNA depletion. Experimental results demonstrate that Ribo-ODDR designed oligos outperform commercially available kits and result in a significant increase in rRNA depletion in Ribo-seq.

A p53-dependent translational program directs tissue-selective phenotypes in a model of ribosomopathies

Gerald C. Tiu et al.

Summary: Ribosomopathies result in tissue-specific phenotypes due to perturbed expression of ribosome components. Mouse genetics research uncovered how p53 and 4E-BP1 play a key role in controlling limb development through selective translational changes. The findings provide an integrative model to understand how tissue-specific phenotypes emerge in ribosomopathies.

Tissue- and stage-specific landscape of the mouse translatome

Hongwei Wang et al.

Summary: The study analyzed the translatomes and transcriptomes of six tissues from mice at embryonic and adult stages, revealing that dynamic translational regulation is mainly achieved through modulation of translational efficiency. Additionally, the research discovered novel microproteins encoded by long noncoding RNAs and demonstrated their regulatory potential and functional relevance.

Widespread translational control regulates retinal development in mouse

Kaining Chen et al.

Summary: The study reveals pervasive translational regulation during retinal development with dynamic changes at the translational level in a developmental stage-specific manner. It also identifies genes whose translational efficiencies are frequently controlled by changes in upstream open reading frame, enriching functions highly important to neurons. Additionally, hundreds of previously uncharacterized micropeptides translated from putative long non-coding RNAs and circular RNAs are discovered, showing potentials in regulating retinal development.
