Full circle: a brief history of cohesin and the regulation of gene expression

Summary: The cohesin complex plays crucial roles in various cellular processes including chromatid cohesion during mitosis, repair of DNA breaks, and control of gene transcription. Recent studies have shown that cohesin depletion has differential effects on gene expression in a cell type- and condition-specific manner. The formation of chromatin loops, mediated by cohesin, is thought to be responsible for its gene regulatory properties, however, a universal mechanism for cohesin-mediated gene expression control remains elusive.

Local and global crosstalk among heterochromatin marks drives DNA methylome patterning in Arabidopsis

The establishment of mCH is dependent on the presence of mCG, and the efficiency of mCH targeting depends on the relative levels of mCG in the genome. The study also proposes a model where local positive feedback and global negative feedback regulate DNA methylation patterning.

Barley Anther and Meiocyte Transcriptome Dynamics in Meiotic Prophase I

Summary: In this study, the temporal dynamics of transcript abundance in barley anthers and meiocytes were investigated using RNA-seq, revealing significant transcriptional changes in anthers at the transition from pre-meiosis to leptotene-zygotene and remarkable stability in meiocytes throughout prophase I after the initial reprogramming at meiosis entry. Additionally, only 24% of putative meiotic gene orthologs showed differential transcript abundance, indicating the complexity of regulatory networks involved in early meiotic processes.

Summary: The molecular origins of spontaneous epialleles are associated with a positive feedback loop between H3K9me2 and CMT2/3. Epialleles, meiotically heritable variations in gene expression, are common in plant genomes and can result from disruptions to heterochromatin methylation maintenance pathways. Feedback regulation of pathways maintaining heterochromatin silencing leads to spontaneous hypermethylated epialleles.

Transgenerational effect of mutants in the RNA-directed DNA methylation pathway on the triploid block in Arabidopsis

Summary: Our study found that the ability of mutants in the RdDM pathway to suppress the triploid block depends on their degree of inbreeding. While first homozygous generation mutants in RdDM components NRPD1, RDR2, NRPE1, and DRM2 have weak or no ability to rescue the triploid block, they are able to suppress the triploid block with successive generations of inbreeding. Inbreeding of nrpd1 was connected with a transgenerational loss of non-CG DNA methylation on sites jointly regulated by CHROMOMETHYLASES 2 and 3.
