Chromatin domain alterations linked to 3D genome organization in a large cohort of schizophrenia and bipolar disorder brains

Kiran Girdhar et al.

Summary: In this study, the researchers constructed chromosomal domains and analyzed histone modifications in the prefrontal cortex of schizophrenia and bipolar disorder patients. They found that there is convergence between genetic risk, neuronal function, and three-dimensional genomics in these disorders. Additionally, they discovered dysregulation of regulatory sequences in the brains of these patients, which are organized into chromosomal domains of various sizes.

Changes in genome architecture and transcriptional dynamics progress independently of sensory experience during post-natal brain development

Longzhi Tan et al.

Summary: The study created a transcriptome and 3D genome atlas of the developing mouse cortex and hippocampus using high-resolution methods, revealing extensive transformations in the transcriptome and 3D genome during early development. The rewiring of the 3D genome in neurons across scales is correlated with gene expression modules and independent of sensory experience.

Cohesin mediates DNA loop extrusion by a swing and clamp mechanism

Benedikt W. Bauer et al.

Summary: The study analyzed how human cohesin-NIPBL complexes mediate loop extrusion, identifying DNA binding sites and large-scale conformational changes required for this process. Results indicate that DNA is translocated by a specific structural element of the cohesin complex to achieve loop extrusion. These findings reveal mechanistic principles of how cohesin-NIPBL and other SMC complexes mediate loop extrusion.

Alterations in Chromatin Structure and Function in the Microglia

Yuki Fujita et al.

Summary: Microglia are resident immune cells in the CNS that exhibit diversity in morphology, density, gene expression profiles, and functions. Recent studies have shown that microglia can alter their gene expression profiles in response to different contexts, such as developmental stages and disease progression. Epigenetic changes, including histone modifications and DNA methylation, play a key role in regulating gene expression and cellular state in microglia, potentially leading to advances in therapeutic approaches for diseases.

Distinct nuclear compartment-associated genome architecture in the developing mammalian brain

Sajad Hamid Ahanger et al.

Summary: This study utilized GO-CaRT technology to investigate genome architecture in neural precursors, revealing the relationship between LAD architecture and regional identity of neural precursor cells, as well as identifying transcriptionally highly active sub-domains within LADs. Additionally, the study found speckle-associated domains to be enriched for genetic loci associated with schizophrenia risk.
