Macro- and Microscale Stress-Associated Alterations in Brain Structure: Translational Link With Depression

Summary: Major depressive disorder (MDD) is a stress-related disorder with cytoarchitectural and neuro-chemical changes that are difficult to detect in the living brain. Research on animal stress models and postmortem human brain tissue has improved understanding of MDD pathophysiology. Evidence from magnetic resonance imaging in humans and animals suggests corticolimbic alterations are linked to depression symptomatology, while cytoarchitectural changes affecting neurons, astroglia, and synapses in MDD are similar to those seen in rodent chronic stress models and are linked to emotion-related behavioral deficits. New approaches using positron emission tomography have been developed to measure synaptic and astroglial alterations in vivo and inform on the contribution of these changes to MDD symptomatology and treatment of stress-related disorders.
