Mitochondria-lysosome contact site dynamics and misregulation in neurodegenerative diseases

Summary: Neurons rely on properly regulated mitochondrial and lysosomal homeostasis, and dysfunction in these two organelles is linked to various neurodegenerative diseases. Recently, mitochondria-lysosome membrane contact sites have been identified as a key pathway mediating their crosstalk in neurons. The regulation of mitochondria-lysosome contact dynamics and its additional functions in regulating organelle network dynamics and metabolite transfer have been highlighted.

Summary: Ischemic stroke, the third leading cause of death in the Western world, mainly affects the elderly and is exacerbated by comorbid conditions such as atherosclerosis or diabetes. Research shows that acute peripheral inflammation worsens stroke-induced neuronal damage in aged mice, linked to increased levels of plasma proinflammatory cytokines. Targeting molecular pathways involved in this process may help improve outcomes for elderly stroke patients.

Intercellular mitochondrial transfer as a means of tissue revitalization

Summary: Mitochondria, crucial for cell metabolism and tissue survival, undergo morphological changes and can be transferred intercellularly. The phenomenon of mitochondrial transfer plays a key role in tissue homeostasis, damaged tissue repair, tumor progression, and immunoregulation, functioning under both physiological and pathological conditions. Various mechanisms contributing to this process, such as trigger factors and transfer routes, are summarized in order to explore potential therapeutic strategies for rescuing tissue damage and inhibiting tumor progression through mitochondria-targeted applications.
