Visualizing T-Cell Responses: The T-Cell PET Imaging Toolbox

Chao Li et al.

Summary: T lymphocytes are crucial regulators in the adaptive immune response, and inappropriate or imbalanced responses can affect the spatiotemporal dynamics and functional status of T cells. Noninvasive PET imaging offers a highly specific and sensitive method to monitor T-cell dynamics in vivo.

CXCR4 expression in glioblastoma tissue and the potential for PET imaging and treatment with [68Ga]Ga-Pentixafor/[177Lu]Lu-Pentixather

Sarah M. Jacobs et al.

Summary: CXCR4 is found to be overexpressed in multiple human cancer types but is low or absent in healthy tissue. In glioblastoma, it is associated with poor prognosis. The study aimed to investigate CXCR4 expression in glioblastoma tissue using PET agents and found a subset of tumors with high CXCR4 expression may be good candidates for targeted radionuclide therapy.

Study of 89Zr-Pembrolizumab PET/CT in Patients With Advanced-Stage Non-Small Cell Lung Cancer

Anna-Larissa N. Niemeijer et al.

Summary: This study investigated the response of NSCLC patients to pembrolizumab monotherapy and the uptake of Zr-89-pembrolizumab in tumors, finding that patients with higher uptake had better responses to treatment. Although no significant correlation was found with PD-L1 or PD-1 immunohistochemistry, the injection of Zr-89-pembrolizumab was deemed safe in the study.

Simpson Grade Revisited - Intraoperative Estimation of the Extent of Resection in Meningiomas Versus Postoperative Somatostatin Receptor Positron Emission Tomography/Computed Tomography and Magnetic Resonance Imaging

Moritz Ueberschaer et al.

Summary: The study evaluated the Simpson grading in relation to modern postoperative imaging techniques and found that intraoperative Simpson grading may underestimate tumor remnants, particularly in grade I and II resections. Postoperative PET-CT showed higher detection rates compared to MRI, indicating the need for further prospective investigation into the prognostic impact of postoperative meningioma remnants.

Assessment of Central Nervous System Lymphoma Based on CXCR4 Expression In Vivo Using 68Ga-Pentixafor PET/MRI

Angelika M. Starzer et al.

Summary: The purpose of this report is to evaluate the feasibility of Ga-68-pentixafor PET for the assessment and follow-up of CNSL. The study showed high accuracy of Ga-68-pentixafor PET in lesion detection, with PTV41% demonstrating the highest concordance with lesion morphology. Future studies should focus on testing the clinical value of Ga-68-pentixafor PET in distinguishing between glioma and CNSL, and between radiation-induced inflammation and viable residual tumor.

Contribution of PET imaging to radiotherapy planning and monitoring in glioma patients - a report of the PET/RANO group

Norbert Galldiks et al.

Summary: Management of patients with glioma usually involves multimodality treatment, with accurate neuroimaging playing a central role in radiotherapy planning and follow-up. The use of PET imaging for radiotherapy planning and monitoring in gliomas has gained interest, but Class I data is currently lacking. PET has potential for response assessment and distinguishing tumor progression from pseudoprogression or radiation necrosis in glioma patients.

Jan-Michael Werner et al.

Summary: The study evaluated the value of FET-PET parameters in diagnosing pseudoprogression related to lomustine-temozolomide chemoradiation, finding that TBRmean had the highest accuracy in identifying pseudoprogression. Furthermore, integrating relative changes of TBRmean improved accuracy, and combining static and dynamic parameters increased specificity to 100%.

Summary: The novel F-18-SiTATE peptide with remarkable imaging properties may help overcome the shortcomings of Ga-68-labeled ligands and foster widespread use of SSTR ligands in imaging for meningioma patients. Compared to Ga-68-labeled ligands, F-18-SiTATE provides important information with comparable quantitative uptake and very good spatial resolution, potentially increasing its utility in clinical practice.

PET Diagnostic Molecules Utilizing Multimeric Cyclic RGD Peptide Analogs for Imaging Integrin αvβ3 Receptors

Christos Liolios et al.

Summary: Multimeric ligands consisting of multiple pharmacophores connected to a single backbone have been widely studied for diagnostic and therapeutic purposes. In targeting integrin alpha(v)beta(3) receptors on cancer cells for molecular imaging and diagnostic applications, multimeric radioligands have shown superiority over monomers. Structural characteristics, binding avidity, pharmacodynamic properties, and PET imaging features of these multimeric ligands have been comprehensively analyzed and discussed in this review.

Imaging of T-cell Responses in the Context of Cancer Immunotherapy

Zebin Xiao et al.

Summary: Immunotherapy is a rapidly expanding approach to treating cancer by enhancing cytotoxic T lymphocytes' function within tumors, but there are still unknown factors in inducing effective anticancer immune responses. Efforts are being made to develop comprehensive ex vivo assessments and noninvasive imaging approaches to detect immune targets and evaluate responses to immunotherapy. These imaging strategies could also be used to monitor treatment responses and predict prognosis for patients treated with immunotherapy.

PSMA Expression in Glioblastoma as a Basis for Theranostic Approaches: A Retrospective, Correlational Panel Study Including Immunohistochemistry, Clinical Parameters and PET Imaging

Adrien Holzgreve et al.

Summary: The study found that PSMA is expressed in glioblastoma both at initial diagnosis and at recurrence. High vascular PSMA expression at recurrence appears to be a negative prognostic marker. Therefore, PSMA expression in GBM could potentially be a promising target for treatment approaches in recurrent glioblastoma.

Summary: PSMA expression in gliomas is associated with neovascularization, with higher expression in high-grade gliomas. Gliomas without IDH1-R132H mutation show significantly more vascular PSMA expression. Additionally, higher PSMA staining vessels are found in tumors with elevated amino acid metabolism, indicating a correlation with shorter survival.
