Ductal Ngn3-expressing progenitors contribute to adult β cell neogenesis in the pancreas

Christopher Gribben et al.

Summary: Ductal cells have been identified as a source of adult beta cell neogenesis, contributing to the beta cell population over time. The expression of Ngn3 in ductal cells plays a role in adult beta cell neogenesis, potentially impacting diabetes onset. Additionally, single-cell RNA sequencing revealed duct-derived somatostatin-expressing cells gave rise to beta cells.

capCLIP: a new tool to probe translational control in human cells through capture and identification of the eIF4E-mRNA interactome

Kirk B. Jensen et al.

Summary: This study introduces a novel method, capCLIP, to comprehensively capture and quantify the eIF4E 'cap-ome' and investigate the biological consequences of eIF4E-cap binding in different cellular contexts. The results from analyzing human cells with mTORC1 inhibitor rapamycin and examining phosphorylation of eIF4E demonstrate that capCLIP is a valuable tool for studying the function of eIF4E and other cap-binding proteins.

Reinforcing one-carbon metabolism via folic acid/Folr1 promotes β-cell differentiation

Christos Karampelias et al.

Summary: Research in zebrafish and neonatal pig islets revealed that overexpression of folate receptor 1 (folr1) and treatment with folinic acid can promote beta-cell differentiation, originating from ductal cells. Comparative metabolomic analysis suggested that changes in pyrimidine, carnitine, and serine pathways may contribute to beta-cell regeneration.

In vivo screen identifies a SIK inhibitor that induces β cell proliferation through a transient UPR

Jeremie Charbord et al.

Summary: A high-throughput chemical screen identified the salt-inducible kinase inhibitor HG-9-91-01 as a driver of beta cell proliferation, acting through an ATF6-dependent unfolded protein response. Mechanistic studies revealed that HG-9-91-01 induces multiple signaling effectors downstream of SIK inhibition, collectively driving beta cell proliferation.

Pre-existing beta cells but not progenitors contribute to new beta cells in the adult pancreas

Huan Zhao et al.

Summary: The study uses genetic lineage tracing to demonstrate that pancreatic endocrine and exocrine progenitor cells do not generate new beta cells, thus arguing against beta-cell neogenesis in the adult mouse pancreas.
