The expanding world of tRNA modifications and their disease relevance

Tsutomu Suzuki

Summary: tRNAs undergo heavy post-transcriptional modifications, which play crucial roles in accurate codon recognition and reading frame maintenance during protein synthesis. Recent studies have shown that tRNA modifications can be dynamically altered in response to cellular and environmental stresses, and deficiencies in tRNA modification can lead to mitochondrial diseases, neurological disorders, and cancer.

Mechanisms and regulation of protein synthesis in mitochondria

Eva Kummer et al.

Summary: Mitochondria are cellular organelles that generate chemical energy through oxidative phosphorylation. They have their own genetic information and unique translation machinery, but also heavily rely on nuclear-encoded proteins and engage in inter-compartmental crosstalk with the cell to regulate their proteome and respond to stress. Understanding the mechanisms and regulation of mitochondrial protein synthesis has increased our knowledge of mitochondrial function and its integration with overall cell physiology.

Targeting Mitochondrial Protein Expression as a Future Approach for Cancer Therapy

Daniela Criscuolo et al.

Summary: Extensive metabolic remodeling is a key feature of cancer cells, with mitochondria playing central roles in cancer development and progression. Coordinated regulation of mitochondrial and cytosolic mRNA translation is crucial for maintaining the integrity of the mitochondrial proteome and functions in cancer cells.

Activation of the integrated stress response confers vulnerability to mitoribosome-targeting antibiotics in melanoma

Roberto Vendramin et al.

Summary: The ability to adapt to environmental stress, including therapeutic insult, plays a role in tumor evolution and drug resistance. The Integrated stress response (ISR) promotes survival by dampening cytosolic translation, but relies on an up-regulation of mitochondrial protein synthesis as well. Targeting mitochondrial ribosomes with antibiotics can sensitize tumors to MAPK inhibition and prevent resistance development.
