Mitochondrial-targeting nanoprodrugs to mutually reinforce metabolic inhibition and autophagy for combating resistant cancer

Min Sun et al.

Summary: This study developed a novel strategy to enhance tumor killing efficacy and combat resistant cancer by mutually reinforcing metabolic inhibition and autophagy. The FG/T-Nanoprodrug, assembled with various functional molecules, demonstrated effective mitochondrial targeting and metabolic inhibition in resistant cancer cells. Through the enhanced chemodynamic therapy, the FG/T-Nanoprodrug was able to induce high levels of autophagy and effectively kill resistant cancer cells in both in vitro and in vivo settings.

Application of glutathione depletion in cancer therapy: Enhanced ROS-based therapy, ferroptosis, and chemotherapy

Boyi Niu et al.

Summary: Glutathione (GSH) plays a crucial role in cellular antioxidative system, particularly in cancer cells, where a high level of GSH is essential for scavenging ROS and detoxifying xenobiotics. Depletion of GSH has been shown to enhance the efficacy of various therapies such as ROS-based therapy, ferroptosis, and chemotherapy. This review comprehensively discusses the importance of GSH depletion in cancer therapy and various strategies employed to achieve it.

Reactive oxygen species (ROS) in cancer pathogenesis and therapy: An update on the role of ROS in anticancer action of benzophenanthridine alkaloids

Abdul Q. Khan et al.

Summary: Reactive oxygen species (ROS) play a crucial role in the growth, progression, and stemness of cancer cells, maintaining a delicate balance with antioxidants to promote cancer pathogenesis. Natural products are vital in attenuating the adverse effects of cancer therapies, with most available drugs derived from natural resources. Benzophenanthridine alkaloids, such as sanguinarine, exhibit pharmacological properties and are being studied for the treatment of various diseases, including cancer, by inducing ROS-mediated anticancer actions.

Remodeling endogenous H2S microenvironment in colon cancer to enhance chemodynamic therapy

Donglin Liu et al.

Summary: Chemodynamic therapy (CDT) is a minimally invasive treatment with high selectivity and promising clinical application, but it lacks effectiveness in inhibiting the growth of colon cancer due to the strongly reducing tumor microenvironment. A new strategy utilizing CuFe2O4 nanoparticles to deplete endogenous H2S and enhance CDT has been proposed, showing significant synergistic effects on improving the performance of CDT for colon cancer.

Lysosomal nitric oxide determines transition from autophagy to ferroptosis after exposure to plasma-activated Ringer's lactate

Li Jiang et al.

Summary: The engineered technology of non-thermal plasma induces ferroptosis and/or apoptosis specifically in cancer cells. Non-thermal plasma-activated Ringer's lactate is another potential modality for cancer therapy. The study found that PAL induces selective ferroptosis in malignant mesothelioma cells through upregulated citrulline-nitric oxide cycle and NF-kappa B activation.
