Making Sense of Nonsense and More: Challenges and Opportunities in the Genetic Code Expansion, in the World of tRNA Modifications

Olubodun Michael Lateef et al.

Summary: The evolution of RNA translation process has been continued through synthetic biology, known as rational bioengineering, which enhances the properties and functionality of proteins. Genetic code expansion allows the incorporation of noncanonical amino acids (ncAAs) into proteins, increasing the range of applications. However, mediating factors reduce the efficiency of ncAA incorporation.

Oligonucleotide Bioconjugation with Bifunctional Palladium Reagents

Muhammad Jbara et al.

Summary: Organometallic reagents have enabled practical strategies for bioconjugation in water, with a focus on peptides, proteins, and oligonucleotides. The development of bifunctional palladium(II)-oxidative addition complexes allows for high-yielding and chemoselective oligonucleotide bioconjugation reactions, even with a broad range of native thiol-containing biomolecules at low micromolar concentrations. This approach provides a versatile method for creating various oligonucleotide conjugates with other biomolecules, such as peptides, proteins, and small molecules, and allows for the conjugation of multiple copies of oligonucleotides onto a monoclonal antibody.

Bioorthogonal information storage in L-DNA with a high-fidelity mirror-image Pfu DNA polymerase

Chuyao Fan et al.

Summary: Researchers have developed a mirror-image DNA that can store genetic information and is resistant to biodegradation, as well as synthesized a high-fidelity mirror-image DNA polymerase. They used this polymerase to encode an article about a mirror-image world in mirror-image DNA, and achieved chiral steganography. The study also showed that mirror-image DNA can be stored in water for longer periods of time, potentially advancing the development of mirror-image biology systems.
