Polymer Pro-Drug Nanoparticles for Sustained Release of Cytotoxic Drugs Evaluated in Patient-Derived Glioblastoma Cell Lines and In Situ Gelling Formulations

Summary: Glioblastoma is the most common and aggressive brain tumor in adults, with limited survival despite multimodal treatments. PEGylated poly(lactide)-poly(carbonate)-doxorubicin nanoparticles showed tumor cell-killing effects, and the release of DOX from polymer-DOX conjugate nanoparticles in thermosensitive matrices was effective over clinically relevant time scales. Future in vivo assessments are needed to evaluate the potential of these nanoparticles in penetrating brain parenchyma.

Correlation between Protonation of Tailor-Made Polypiperazines and Endosomal Escape for Cytosolic Protein Delivery

Summary: Responsive polymers that become protonated at decreasing pH are considered important in the development of synthetic cell entry vectors. This study presents a series of hydrophilic polypiperazines with varying amino groups, allowing for fine-tuning of basicity within the physiologically relevant pH range. The research reveals that polymers with pKa values above 6.5 or 7 can lead to rapid endosomal release in different media, but higher pKa values (>7) can result in cytotoxic effects.
