Dihydroartemisinin as a potential drug candidate for cancer therapy: a structural-based virtual screening for multitarget profiling

Ibrahim Malami et al.

Summary: This study evaluated the binding properties of dihydroartemisinin to multiple protein targets implicated in cancer pathogenesis using molecular docking simulation techniques. The results showed that dihydroartemisinin exhibited excellent binding to various protein targets, with 75% being protein kinases and 25% being non-protein kinases. The findings provide insights for the development of effective anticancer agents.

Summary: CBD shows inhibitory effects on excessive inflammation in a murine model of multiple sclerosis through various mechanisms, such as suppressing the expression of inflammatory mediators, regulating inflammatory macrophage activity, promoting myeloid-derived suppressor cell expansion, and limiting inflammation in the gastrointestinal tract. These findings demonstrate the beneficial impact of CBD treatment on autoimmune neuroinflammation.

Summary: CBD, isolated from cannabis sativa in 1940, has been proven to have anti-seizure effects with no psychoactive activity. Recent clinical trials have confirmed its effectiveness in drug-resistant epilepsy, with Epidiolex (R) being the only cannabis-derived drug approved for epilepsy. Future studies are needed to explore its potential in specific types of epilepsy.

Prophylactic Administration of Cannabidiol Reduces Microglial Inflammatory Response to Kainate-Induced Seizures and Neurogenesis

Tanya R. Victor et al.

Summary: The study examines the effects of commercially available CBD on microglia inflammatory activation and neurogenic response in the presence and absence of seizures. CBD significantly dampens microglial migration and accumulation to the hippocampus, reducing the number of ectopic neurons deposited in the hippocampal area post seizure.

Carlos Villarroel-Vicente et al.

Summary: Natural products and synthetic analogs are potential therapeutic drugs for metabolic syndrome by activating the transcriptional activity of PPARs and managing obesity, diabetes, dyslipidemia, and non-fatty liver disease. Compounds like quercetin, resveratrol, and curcumin have shown beneficial effects on patients. Further large-scale clinical trials may be needed for promising compounds.

Summary: CBD has shown therapeutic potential in seizure disorders, with progress made in understanding its mechanisms of action and clinical properties. A pharmaceutical-grade formulation of purified CBD has been approved for treatment in certain seizure syndromes, but interactions with other medications, such as clobazam, remain a complex issue. Despite advances, there are still significant gaps in knowledge regarding CBD's antiseizure activity.

Neuroprotective versus Neuroinflammatory Roles of Complement: From Development to Disease

Marlene Kanmogne et al.

Summary: Complement proteins play important roles in innate immunity and regulation of neural networks in the central nervous system. While classical complement proteins are implicated in CNS dysfunction, recent data also suggest neuroprotective roles in CNS homeostasis through refinement of synaptic connections. Understanding these mechanisms may lead to novel treatment targets for CNS diseases involving complement-mediated synapse loss.

Cannabidiol-dihydroartemisinin conjugates for ameliorating neuroinflammation with reduced cytotoxicity

Fanfan Wang et al.

Summary: The study found that the combination of CBD and DHA has an additive effect in inhibiting LPS-induced nitric oxide production, but synergizes in inhibiting BV-2 cellular viability, suggesting an amplification of cytotoxicity. The CBD-DHA conjugate C3D eliminated cytotoxicity associated with single CBD/DHA use without compromising anti-neuroinflammation activity. C3D was more potent than C2D and C4D in inhibiting LPS-induced NO and mRNAs of iNOS and IL-1 beta, indicating that linker length is critical for the anti-inflammatory activities of CBD-DHA conjugates.

However, the bidirectional mechanism of the reciprocal interaction between epilepsy and neuroinflammation still needs further understanding.
