Systematic Phenotyping and Characterization of the 3xTg-AD Mouse Model of Alzheimer's Disease

Summary: Animal models are important for studying disease mechanisms and potential treatments, but the availability of numerous animal models presents challenges for Alzheimer's disease (AD) researchers. In this study, the 3xTg-AD mouse model was analyzed to understand the specific pathologies that develop at different ages and to comment on changes since its development 20 years ago. The study also compared the 3xTg-AD model with the 5xFAD model and found that plasma NfL is influenced by plaque burden. The results are freely available for researchers and demonstrate the usefulness of a standardized characterization pipeline for investigating and comparing different AD models.

[18F]Nifene PET/CT Imaging in Mice: Improved Methods and Preliminary Studies of α4β2*Nicotinic Acetylcholinergic Receptors in Transgenic A53T Mouse Model of α-Synucleinopathy and Post-Mortem Human Parkinson's Disease

Summary: This study reported the binding of [F-18]nifene to nAChRs in Parkinson's disease using a transgenic mouse model and postmortem human brain sections. Improved radiosynthesis methods and PET/CT procedures in mice were developed to establish optimal imaging time and observe decreases in [F-18]nifene binding both in vivo and in vitro. Confirmation of Lewy bodies and alpha-synuclein aggregates in human PD brain sections further supported the potential of [F-18]nifene as a valuable tool for PET imaging studies of PD.

[18F]FDG PET/CT Studies in Transgenic Hualpha-Syn (A53T) Parkinson's Disease Mouse Model of α-Synucleinopathy

Summary: The M83 transgenic mouse line is a model of alpha-synucleinopathy found in Parkinson's disease, useful for studying non-motor deficits in the early stages of PD. Research findings indicate metabolic changes and neurodegenerative symptoms in this PD mouse model, leading to progressive motor dysfunction and death. 9-month-old male and female Hualpha-Syn (A53) mice exhibit up to 30% decrease in [F-18]FDG uptake, consistent with cortical hypometabolism observed in PD patients.
