Amorphous Drug-Polymer Salt with High Stability under Tropical Conditions and Fast Dissolution: The Case of Clofazimine and Poly(acrylic acid)

Summary: The stability of amorphous clofazimine against crystallization is significantly improved by forming a salt with a polymer, without sacrificing dissolution rate. The amorphous salt at high drug loading shows remarkable stability under harsh conditions and fast dissolution in biorelevant media. This study suggests a general method for preparing amorphous drugs with high physical stability and fast dissolution.
