Optimization of AsCas12a for combinatorial genetic screens in human cells

Peter C. DeWeirdt et al.

Summary: In this study, an optimized Cas12a toolkit was developed for pooled, combinatorial genetic screens in human cells. By assaying the activity of thousands of guides, refined on-target design rules and comprehensive off-target rules were established. The toolkit was validated by screening for synthetic lethality in cancer cells and showed similar performance to Cas9 in genome-wide dropout screens with greatly reduced library size.

Mapping the genetic landscape of DNA double-strand break repair

Jeffrey A. Hussmann et al.

Summary: Cells repair DNA double-strand breaks using a complex set of pathways, with the study identifying unexpected relationships among DSB repair genes and demonstrating the importance of genetic dependencies in repair outcomes. The research lays a foundation for mapping DNA repair pathways and optimizing genome editing across a variety of modalities.

Single-cell lineages reveal the rates, routes, and drivers of metastasis in cancer xenografts

Jeffrey J. Quinn et al.

Summary: This study used a Cas9-based lineage tracer to track the development and metastasis of lung cancer cells in a mouse model over months, revealing heterogeneity in metastatic capacity and genetic differences that drive invasiveness. The study also uncovered some genes driving metastasis and an unexpected suppressive role of KRT17 in metastasis.

Recording of elapsed time and temporal information about biological events using Cas9

Jihye Park et al.

Summary: The study discovered that indel generation by Cas9 and guide RNA can be used to measure absolute time periods in mammalian cells. The method was successfully applied in various cell types and living mice to record the duration of chemical exposure and the elapsed time since the onset of biological events. The system proposed in the study could potentially serve as synthetic DNA clocks.

A cis-acting mechanism mediates transcriptional memory at Polycomb target genes in mammals

Daniel Holoch et al.

Summary: Epigenetic inheritance plays a role in maintaining cellular identities and transcriptional memory, particularly at Polycomb targets, depends on both histone modifications and the gene-regulatory network, varying with cellular context. Polycomb proteins are involved in regulating this type of epigenetic transcriptional memory during cell differentiation.

Integrated spatial genomics reveals global architecture of single nuclei

Yodai Takei et al.

Summary: This study utilized DNA seqFISH+ technology to image 3,660 chromosomal loci in single mouse embryonic stem cells, along with observing 17 chromatin marks and subnuclear structures by sequential immunofluorescence and the expression profile of 70 RNAs. The results suggest that many loci are consistently associated with immunofluorescence marks and form "fixed points" in nuclear organizations in single cells, while highly expressed genes appear to be pre-positioned to active nuclear zones. The research also uncovered distinct mouse ES cell subpopulations with characteristic combinatorial chromatin states.

Bimodal regulation of the PRC2 complex by USP7 underlies tumorigenesis

Dongxue Su et al.

Summary: This study reveals the regulation of EZH2 abundance and function by USP7 in cancer cells, highlighting the potential of USP7 as a therapeutic target for cancer intervention. The physical association between USP7 and PRC2 complex plays a critical role in cell growth, proliferation, and tumorigenesis, with implications for histological grades and prognosis of tumor patients.
