Multi-dimensional fragmentomic assay for ultrasensitive early detection of colorectal advanced adenoma and adenocarcinoma

Xiaoji Ma et al.

Summary: An integrated model using fragmentomic profiles of plasma cell-free DNA was established for accurately detecting early-stage CRC and advCRA. The model demonstrated high sensitivity and specificity in an independent test cohort, showing particularly strong performance in identifying early-stage CRC.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Genome-wide cell-free DNA methylation analyses improve accuracy of non-invasive diagnostic imaging for early-stage breast cancer

Jiaqi Liu et al.

Summary: This study demonstrated the clinical utility of cfDNA methylation markers in improving the early diagnosis of BC patients in BI-RADS category-4, significantly increasing the accuracy; the results suggest that combining liquid biopsy with traditional diagnostic imaging can enhance the detection of breast cancer.

Epigenetics, fragmentomics, and topology of cell-free DNA in liquid biopsies

Y. M. Dennis Lo et al.

Summary: Liquid biopsies, which analyze cell-free DNA in blood plasma, are utilized for noninvasive prenatal testing, oncology, and organ transplant monitoring. The physical and molecular features of cell-free DNA fragments, along with their distribution over the genome, contain information about their tissues of origin. These features, including DNA methylation patterns and nucleosomal organization, hold promise for noninvasive detection of tissue-specific pathologies with a variety of clinical applications.

Detection and characterization of lung cancer using cell-free DNA fragmentomes

Dimitrios Mathios et al.

Summary: Non-invasive assessment of cell-free DNA (cfDNA) provides an opportunity for cancer detection and intervention. Using a machine learning model and genome-wide analysis, the study successfully detected tumor-derived cfDNA in a prospective study of 365 individuals at risk for lung cancer. Combining clinical factors and CEA levels, the approach showed high accuracy in distinguishing lung cancer subtypes and the fragmentation score served as an independent prognostic indicator of survival.
