The emerging roles of circRNAs in cancer and oncology

Summary: Circular RNAs (circRNAs) are a novel class of primarily non-coding RNAs with increasingly recognized roles in cancer development and progression through diverse mechanisms of action. In this review, the authors summarize the current understanding of circRNA biogenesis, regulation, physiological functions and pathophysiological roles in cancer. They also discuss the clinical potential of circRNAs as biomarkers, therapeutic agents and drug targets in oncology, as well as the research controversies, technical issues, and biological knowledge gaps that need to be addressed.

PSMC2 promotes the progression of gastric cancer via induction of RPS15A/mTOR pathway

Summary: Gastric cancer is a common malignant tumor and understanding its development mechanism and finding effective therapeutic target genes are crucial. This study found that PSMC2 and RPS15A were highly expressed in gastric cancer. PSMC2 enhanced RPS15A levels by targeting hsa-let-7c-3p, which then activated the mTOR pathway, promoting the progression of gastric cancer.

Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Summary: The study found that CREB1 plays a key role in maintaining luminal cell survival and that oncogenesis changes the CREB1-induced transcriptome. ING4 and JFK were identified as transcriptional targets of CREB1 in luminal cells. Oncogenic suppression of PTEN results in constitutive activation of CREB1, blocking CREB1 suppresses tumor growth. This is the first study to define the molecular mechanism where loss of PTEN in cancer leads to aberrant CREB1 activation and suppression of ING4 expression impacting luminal cell differentiation.
