Structural coordination between active sites of a CRISPR reverse transcriptase-integrase complex

Summary: CRISPR-Cas systems provide adaptive immunity in bacteria and archaea through integration of foreign sequences, with some systems utilizing a single protein for sequence integration and crRNA production. Structural analysis of a Cas6-RT-Cas1-Cas2 complex reveals coordination between active sites and the ability to acquire CRISPR sequences from RNA and DNA substrates. These findings support a model of sequential enzymatic activities in CRISPR systems.
