Spatial Proteomics Reveals Differences in the Cellular Architecture of Antibody-Producing CHO and Plasma Cell-Derived Cells

Comparative systeomics to elucidate physiological differences between CHO and SP2/0 cell lines

Summary: Omics-based tools were used to analyze two biopharma cell types, M-CHO and CHO-K1, and SP2/0, revealing differences in gene expression and protein profiles. M-CHO showed downregulation in genes related to protein folding, protein synthesis, and protein metabolism, while SP2/0 showed reduced expression in cell cycle and actin cytoskeleton genes. Pathway analysis indicated reduced glycerolipid, glycosphingolipid, ABC transporter, calcium signaling, cell adhesion, and secretion pathways in M-CHO, while retinol metabolism was upregulated. The analysis also revealed enrichment of apoptosis, RNA degradation, and proteosome pathways in CHO stationary phase. Membrane proteins and secretory granules were underrepresented in M-CHO. The findings suggest potential opportunities for improving production capabilities of CHO and SP2/0 cells.
