In-Depth Site-Specific O-Glycosylation Analysis of Glycoproteins and Endogenous Peptides in Cerebrospinal Fluid (CSF) from Healthy Individuals, Mild Cognitive Impairment (MCI), and Alzheimer's Disease (AD) Patients

Zhengwei Chen et al.

Summary: Site-specific O-glycoproteome mapping in complex biological systems provides a molecular basis for understanding the structure-function relationships of glycoproteins and their roles in physiological and pathological processes. A comprehensive strategy was developed to analyze the O-glycoproteome in cerebrospinal fluid (CSF) and identify O-glycopeptides from both sialylated and nonsialylated glycoforms. The study also revealed that endogenous peptides in CSF were predominantly O-glycosylated, with a decreasing trend in fucosylation and an increasing trend in endogenous peptide O-glycosylation observed in patients with Alzheimer's disease.

Matrisome changes in Parkinson's disease

Margaret Downs et al.

Summary: In this study, the matrisome proteins in Parkinson's disease (PD) were analyzed and significant differences were found in the expression of collagen proteins compared to control samples. Histological analysis supported these findings and showed similar trends in collagen abundance. The hydroxyprolination of collagen was less variable in PD compared to controls, and glycoproteomic changes were observed. Network topology-based analysis revealed the involvement of angiogenesis-related proteins in PD. Further studies with a larger cohort are needed to validate these results.

In-Depth Matrisome and Glycoproteomic Analysis of Human Brain Glioblastoma Versus Control Tissue

Manveen K. Sethi et al.

Summary: This study investigates the glycosylation of extracellular matrix components in the development of Glioblastoma (GBM). The findings suggest that alterations in GAGs and PGs are closely related to GBM tumorigenesis. The study reveals significant differences in sulfation and glycosylation levels between GBM and control samples, as well as differential regulation of core matrisome proteins. These findings provide important insights into the molecular changes associated with GBM development.

Molecular signature of extracellular matrix pathology in schizophrenia

Harry Pantazopoulos et al.

Summary: Growing evidence suggests that the extracellular matrix plays a critical role in the pathophysiology of schizophrenia, with changes in PNNs and CSPGs identified in glial cells. Gene expression profiling showed widespread dysregulation of ECM gene expression in cortical and subcortical brain regions in SZ, affecting key ECM components. This contributes to emerging evidence of large-scale ECM dysregulation in SZ and highlights molecular pathways involved in PNN decreases and cognitive impairment.
