Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

ACSL family: The regulatory mechanisms and therapeutic implications in cancer

Jing Quan et al.

Summary: The deregulation of fatty acid metabolism is associated with cancer development, with ACSL family members playing different roles in cancer progression, some promoting cancer and some having inhibitory effects, while also participating in cell death processes. ACSLs may serve as important therapeutic targets and biomarkers for cancer treatment.

Emerging roles of dehydrogenase/reductase member 2 (DHRS2) in the pathology of disease

Zhenzhen Li et al.

Summary: DHRS2 is a carbonyl reductase that plays critical roles in regulating the cell cycle, lipid metabolism, and hormone metabolism. Recent studies have shown that DHRS2 can impact cancer cell proliferation, migration, and invasion by reprogramming lipid metabolism and redox homeostasis.

Metabolic Reprogramming of Mammary Epithelial Cells during TGF-β-Induced Epithelial-to-Mesenchymal Transition

Wan Hua et al.

Summary: TGF-beta induces metabolic reprogramming in epithelial cells during EMT, affecting glycolysis, TCA cycle, glutaminolysis, choline metabolism, cellular redox state, and other pathways. Inhibiting choline kinase alpha with a kinase inhibitor can attenuate TGF-beta-induced changes associated with EMT.
