A guide to machine learning for biologists

Joe G. Greener et al.

Summary: This passage discusses the application of machine learning in the analysis of biological data and provides guidance for experimentalists. The increasing scale and complexity of biological data have led to a growing use of machine learning in biology.

Effect of ibrutinib with R-CHOP chemotherapy in genetic subtypes of DLBCL

Wyndham H. Wilson et al.

Summary: In younger patients with non-GCB DLBCL, the addition of ibrutinib to R-CHOP chemotherapy improves survival rates, particularly in patients with MCD and N1 subtypes. This study provides mechanistic understanding of the benefit of ibrutinib in this patient population.

ROBUST: A Phase III Study of Lenalidomide Plus R-CHOP Versus Placebo Plus R-CHOP in Previously Untreated Patients With ABC-Type Diffuse Large B-Cell Lymphoma

Grzegorz S. Nowakowski et al.

Summary: The ROBUST study aimed to compare the efficacy of lenalidomide plus R-CHOP with placebo/R-CHOP in previously untreated ABC-type DLBCL. While the primary end point of progression-free survival (PFS) was not met in all patients, there were trends favoring R2-CHOP over placebo/R-CHOP in patients with higher-risk disease. The safety profile of R2-CHOP was consistent with individual treatments with no new safety signals.

Mutational mechanisms shaping the coding and noncoding genome of germinal center derived B-cell lymphomas

Daniel Huebschmann et al.

Summary: The study revealed distinct mutational signatures in germinal center derived B-cell lymphomas linked to somatic hypermutation and class-switch recombination. Mutations not only affect immunoglobulin genes, but also cause off-target mutations in non-IG genes. B cell-specific mutational processes contribute to both lymphoma initiation and intratumoral heterogeneity through diverse mechanisms.

Engineering large-scale chromosomal deletions by CRISPR-Cas9

Thomas F. Eleveld et al.

Summary: Large-scale chromosomal deletions are common in cancer and confer an oncogenic advantage, but the oncogenic drivers behind these deletions are difficult to identify. This study introduced a novel CRISPR-Cas9 technique to engineer large-scale deletions and create isogenic cell line models, successfully inducing deletions in neuroblastoma cell lines. The technique may provide valuable insights into the role of large-scale deletions in tumor development and growth.
