RNA Sequencing Reveals Cancer-Associated Changes in Laryngeal Cells Exposed to Non-Acid Pepsin

Tina L. Samuels et al.

Summary: In this in vitro study, brief exposure to pepsin was found to activate cancer-associated signaling pathways in laryngeal cells, revealing new mechanisms by which chronic reflux may contribute to carcinogenesis. The developed cell line provides a novel tool to investigate pepsin-dysregulated pathways identified by RNA sequencing and disparities in tumor susceptibility of laryngeal subsites.

Impact of bile acids on the severity of laryngo-pharyngeal reflux

Eugenio De Corso et al.

Summary: This study found that high concentrations of bile acids are associated with the severity of LPR, as well as an increased risk of esophagitis and upper airway malignancies. Salivary bile acids >1 µmol/L may be a reliable indicator of LPR severity, and bile acids were the most suitable molecules for LPR diagnosis, with a sensitivity of 86% and a positive predictive value of 80.7%.

Weakly Acidic Bile Is a Risk Factor for Hypopharyngeal Carcinogenesis Evidenced by DNA Damage, Antiapoptotic Function, and Premalignant Dysplastic Lesions In Vivo

Clarence T. Sasaki et al.

Summary: Recent research has shown that weakly acidic bile can also be carcinogenic in vivo, promoting premalignant lesions in the hypopharynx with oxidative damage and other cellular changes. Antacid therapy may have limited effect on reducing the risk of bile-related hypopharyngeal neoplasia.
